摘要
目的探讨冬凌草甲素对肿瘤转移相关酶(乙酰肝素酶,MMP-2,MMP-9)的影响。方法利用半定量RT-PCR检测乳腺癌MDA-MB-231细胞乙酰肝素酶基因的表达变化,用明胶酶谱检测冬凌草甲素对人乳腺癌MDA-MB-231细胞MMP-2、MMP-9活性的影响。结果冬凌草甲素可下调乙酰肝素酶在乳腺癌MDA-MB-231细胞中的表达,并抑制基质金属蛋白酶MMP-2和MMP-9的活性。结论冬凌草甲素可通过下调乙酰肝素酶基因表达、抑制MMP-2和MMP-9活性,从而抑制肿瘤的侵袭转移。
Objective To observe the influence of oridonin on tumor metastasis-related enzyme( mainly Heparanase, MMP-2, MMP-9). Methods Detect the changes of gene expression of heparanase in breast cancer MDA-MB-231 cells by semi-quantitative RT-PCR, and the activities of MMP-2 and MMP-9 by gelatinase spectrum in human breast cancer MDA-MB-231 treated by oridonin. Result Oridonin could down-regulate the expression of heparanase in breast cancer MDA-MB-231 cells, and inhibit the activities of matrix metalloproteinase( MMP-2, MMP-9). Conclusion Oridonin can down-regulate the expression of heparanase and inhibit the activities of matrix metalloproteinase( MMP-2, MMP-9) to prevent progression and metastasis of tumor.
