彗星试验检测雄黄对中国仓鼠肺细胞DNA的损伤作用

Impairing Effects on DNA in Chinese Hamster Lung Cells Induced by Realgar

目的检测含砷中药雄黄对中国仓鼠肺细胞(CHL细胞)DNA的损伤作用。方法测定雄黄对CHL细胞的半数抑制浓度(IC50),根据IC50设立不同剂量组,进行彗星试验,观察并比较不同组别不同损伤等级细胞数的差异。结果给药干预后,各剂量组及阳性对照组DNA损伤细胞数明显增加,且各剂量组随着给药浓度的增加,Ⅲ、Ⅳ级(中、重度)损伤细胞数增多。结论中药雄黄可引起CHL细胞DNA损伤。

Objective To explore the damage on DNA of Realgar in CHL cells. Methods The IC50 of Realgar on CHL cells was determined to establish the scope of doses, and comet assay was done to observe and compare the difference within the groups with different doses and with different damage ranks. Results The number of DNA damage cells was increased significiantly in all dose groups and positive group; and with the dose increase,the number of Ⅲand Ⅳ of DNA damage ranks increased. Conclusion Chinese medicine Realgar can induce DNA damage on CHL cells.
