缺氧诱导因子1α、葡萄糖转运蛋白1在宫颈癌中的表达及二者相关性研究 被引量:1

The Expression of Hypoxia Inducible Factor-1α and Glucose Transporter-1 in Cervical Cancer and Their Associations with Carcinogenesis

摘要 目的检测宫颈癌中缺氧诱导因子1α(HIF-1α)、葡萄糖转运蛋白1(GLUT-1)的表达,探讨二者的相关性及临床意义。方法应用免疫组化法检测HIF-1α及GLUT-1的表达情况。结果HIF-1α、GLUT-1在宫颈癌和宫颈上皮内瘤变(CIN)中的表达明显高于正常宫颈上皮;宫颈癌中,HIF-1α表达与患者年龄、肿瘤大小、病理类型及分化程度无相关性,而与淋巴结转移情况及临床分期相关(P<0.05);GLUT-1表达与分化程度、淋巴结转移情况及临床分期相关(P<0.05),与患者年龄、肿瘤大小及病理类型无相关性;HIF-1α、GLUT-1表达水平呈正相关。结论HIF-1α、GLUT-1的过度表达可能与宫颈癌的发生、发展有关,且HIF-1α可能提高GLUT-1的表达。HIF-1α、GLUT-1有望成为宫颈癌诊断的早期预测指标及基因治疗的新靶点。 Objective To investigate the expression of HIF-1α and GLUT-1 in cervical cancer and to study their relationships and significance in clinical research. Methods The expression of HIF-1α and GLUT-1 was detected by immunohistochemistry. Results The expression rates of HIF-1α and GLUT-1 in cervical cancer and CIN (cervical intraepithelial neoplasia)were significantly higher than those in normal cervical tissues and CIN. In cervical cancer, HIF -1α expression had no relationship with age, tumor size, pathologic type or histological differentiation, but with disease stage and pelvic lympt mode metastasis status. The expression of GLUT-1 was related with histological differentiation, pelvic lymph mode metastasis and disease stage, but not with age, tumor size and pathologic type. There were positive correlations between the expression of HIF-1α and GLUT-1. Conclusion Overexpression of HIF-1α and GLUT-1 is associated with the carcinogenesis and development of cervical cancer,the expression of GLUT-1 is enhanced by HIF-1α. HIF-1α and GLUT-1 may be hoped as biological markers for cervical cancer diagnosis and new gene therapy targets.
