重组人干扰素-γ对腭裂术后裸露骨面瘢痕成纤维细胞的影响 被引量:2

摘要 目的探讨重组人干扰素γ(rhIFN-γ)对体外培养的腭裂术后裸露骨面瘢痕来源的成纤维细胞生物学作用的影响。方法制作兔腭裂术后裸露骨面瘢痕动物模型,将rhIFN-γ加入瘢痕成纤维细胞培养基内,采用MTT法测定细胞增殖率,应用流式细胞仪分析rhIFN-γ对腭裂瘢痕成纤维细胞的细胞周期生物学行为的影响;采用SPSS13.0软件对数据进行分析。结果和对照组比较,实验组加入rhIFN-γ后,腭裂术后裸露骨面瘢痕成纤维细胞增殖活性及DNA合成被明显抑制,S-G2-M期细胞百分数明显降低。结论rhIFN-γ可以抑制腭裂术后裸露骨面瘢痕成纤维细胞的DNA合成及增殖,其与细胞数量改变之间有一定的剂量-效应关系,提示rhIFN-γ是成纤维细胞负性调节因子,在防治腭裂术后裸露骨面瘢痕的形成中有一定的应用价值。 Objective To observe the biological effects of recombinant human interferon-γ(rhIFN-γ) on rabbit palatal scar fibroblasts in vitro. Methods Establish the rabbit model of cleft palate bone denudation after surgery. RhIFNγ/was added to the cultivated hypertrophic scar fibroblasts. MTT assay was used to test the effects of rhIFN-γ on proliferation of scar fibroblasts. Flow cytometry was also employed to measure the influence of rhIFN-γ to the biological behavior of the palate scar fibroblast cell division cycle. SPSS 13.0 software package was used to analyze the data. Results Compared with the control group, after cleft palate surgery, the proliferation of scar fibroblasts and DNA synthesis were inhibited, and cells were significantly lower percentage at the S-G2-M phase in the experimental group. Conclusion RhIFN-γcould inhibit DNA synthesis and proliferation of scar fibroblasts after the surgery of cleft palate bone denudation with a certain amount of change between the dose-response relationship, suggesting that rhIFN-γ is the negative regulator of fibroblasts, which have a certain value in the prevention and treatment of cleft palate bone denudation of the scar formation.
