摘要
目的观察芪参益气滴丸对急性心肌梗塞(AMI)大鼠不同时间点心功能的影响。方法制备AMI大鼠,随机分成假手术组、AMI模型组、芪参益气滴丸治疗组[3.5 mg/(100 g.d)芪参益气滴丸混悬液灌胃给药]、卡托普利治疗组[1.0125 mg/(100 g.d)卡托普利悬浊液灌胃给药],每组又分4个时点(3 d,2周,4周,8周),共16个组,每组9只。于各时间点进行超声心动检查。结果AMI组与假手术组相比,各时点LVDd,LVEDV,E/A值显著增加(P<0.05);LVEF,FS%,Wti显著降低(P<0.05),LVPWD 3 d,2周,4周时显著降低(P<0.05),8周时显著增高(P<0.05)。芪参益气滴丸治疗组与AMI组比较,2周,4周,8周时LVDd,LVEDV显著降低(P<0.05);4周时LVPWD,2周,4周,8周时WTi显著升高(P<0.05);8周时LVPWD显著降低(P<0.05);4周,8周LVEF,2周,4周,8周FS%显著升高(P<0.05);2周,4周,8周时E/A值显著减低(P<0.05)。卡托普利治疗组与AMI组比较,在4周,8周时,LVDd、LVEDV显著降低(P<0.05);4周时LVPWD显著升高(P<0.05);8周时LVPWD显著降低(P<0.05);4周,8周LVEF,2周,4周,8周FS%显著升高(P<0.05);2周,4周,8周时E/A值显著减低(P<0.05)。芪参益气滴丸治疗组与卡托普利治疗组比较,2周,4周,8周时较卡托普利治疗组降低LVDd,LVEDV作用明显,两组有显著差异(P<0.05),4周,8周时芪参益气滴丸治疗组Wti变薄程度减轻,FS%升高明显,两组均有显著差异(P<0.05)。结论芪参益气滴丸早期应用具有改善AMI大鼠心功能的作用。
Objective To observe the effects of Qishenyiqi Dripping Pills on the expression of TNF - α mRNA in rats with acute myocardial infarction(AMI) at different time point. Methods Myocardial infarction model was replicated by ligating left anterior descending branch of coronary artery in wistar rats. Rats were randomly divided into sham operation group, AMI group, Qishenyiqi Dripping Pill group, Captopril group , and every group was divided into 4 different time points ( 3 days,2 weeks ,4weeks, 8 weeks ). Animals underwent euthanasia at the end of the study, and the level of TNF - α mRNA in no infarct and infract area of left ventricle was examined by RT - PCR. Results The mRNA expression of TNF - α in AMI group were significantly elevated as compared with sham rats ( P 〈 0.05 ). TNF - α in Qishenyiqi Dripping Pill group and Captopril group was markedly attenuated as compared with MI group and there were significant different in week 2, week 4 and week 8 ( P 〈 0.05 ). TNF - α in infract area of Qishenyiqi Dripping Pill group was markedly attenuated as compared with Captopril group ( P 〈 0.05 ). TNF - α in no infarct area of Qishe- nyiqi Dripping Pill group was markedly attenuated compared with Captopril group on day 3 and in week 2 ( P 〈 0.05 ). Conclusion Qishenyiqi Dripping Pills can inhibit inflammatory reaction after acute myocardial infarction, relieve degree of myocardial injury, so as to protect cardiac function.
