何首乌饮对衰老大鼠肾脏增殖与凋亡的影响 被引量:5

摘要 目的研究中药何首乌饮对衰老大鼠肾脏增殖及凋亡的影响。方法50只SD大鼠随机分为5组:阴性对照组,模型组,何首乌饮高、中、低剂量组,每组10只。D-半乳糖连续腹腔注射制作亚急性衰老大鼠模型。采用SP免疫组化法和原位末端标记法(TUNEL法)分别检测肾脏组织PCNA和凋亡的表达情况。结果①模型组大鼠肾脏PCNA阳性细胞数低于阴性对照组大鼠(P<0.05)。何首乌饮各剂量组PCNA阳性细胞数高于模型组(P<0.05)。②模型组大鼠肾脏TUNEL阳性细胞数高于阴性对照组大鼠(P<0.05)。何首乌饮各剂量组TUNEL阳性细胞数均低于模型组(P<0.05)。结论何首乌饮可明显提高衰老大鼠肾脏的增殖能力并抑制其凋亡,具有一定的延缓衰老作用。 Objective To observe the effects of traditional Chinese medicine Heshouwuyin on the expression of PCNA and apoptosis in kidney of aging rats. Methods 50 SD rats were randomly derided into 5 groups : negative control group, aging model group, Heshouwuyin groups( high,middle and low dose groups) , and each group had 10 rats. The sub - acute aging model rats were made by injecting D - gal into abdounmal cavity continuously. SP immunohistochemistry and Tdt - mediated dUTP nick end labelling (TUNEL) were used to observe the expression of PCNA and apoptosis in kidney. Results ①The counts of PCNA positive cells of model group were lower than negative control group ( P 〈 0. 05 ). Those of Heshouwuyin groups were higher than model group ( P 〈 0.05 ). ②The counts of apoptotic ceils of model group were higher than negative control group ( P 〈 0.05 ). Conclusion Traditional Chinese medicine Heshouwuyin can increase PCNA expression and decrease apoptosis in kidney of aging rats obviously, and has fairly anti - aging effects.
