摘要
目的研究姜黄素对P糖蛋白(P-gp)介导的膀胱肿瘤多药耐药的逆转作用。方法采用MTT法测定药物的体外杀伤作用,应用流式细胞术测定细胞内罗丹明浓度。结果姜黄素不增加阿霉素对BIU-87的细胞毒性作用(P>0.05);而对于BIU-87/ADR,姜黄素明显增加阿霉素细胞毒性(P<0.05),逆转指数为4.0。姜黄素明显抑制P-gp的药物外排作用可能是关键因素。结论姜黄素通过抑制P-gp的药物外排作用有效逆转多药耐药。
Objective To investigate the reversal effect on P -gp mediated bladder tumor muhidrug resistance(MDR) by curcumin. Methods The reversal effect of curcumin on muhidrug - resistance was determined by MTT method. The drug efflux function of P - glycoprotein was studied by flow cytometry. Results The cytotoxicity of curcumin plus adriamycin and free adriamycin was identical in BIU - 87 cells, but the chemosensitivity of BIU - 87/ADR cells to curcumin plus adriamycin was increased to 4.0 times compared with that of BIU - 87/ADR cells to free adriamycin. Curcumin obviously inhibited the drug efflux function of P - glycoprotein maybe a key factor. Conclusion Curcumin reverses MDR effectively by inhibiting the drug efflux function of P - glyeoprotein.
