短发夹状RNA抑制子宫颈癌Siha细胞HPV16 E6基因的表达

摘要 目的研究人乳头状瘤病毒(HPV)16 E6 siRNA表达载体对宫颈癌Siha细胞株HPV16 E6基因的抑制作用。方法利用脂质体将HPV16E6特异性si RNA表达载体转染宫颈癌Siha细胞株,应用荧光定量RT-PCR检测HPV16 E6 mRNA的变化,流式细胞术检测E6蛋白表达的变化。结果细胞试验表明HPV16 E6siRNA表达载体可明显抑制Siha细胞E6 mRNA和E6蛋白的表达,抑制率分别为92.65%和84.40%。且其抑制效果可维持4周以上。结论HPV16 E6 siRNA表达载体可以高效、特异、长期的抑制宫颈癌Siha细胞HPV16 E6基因的表达。 Objective To observe the inhibitory effects of short hairpin RNA on the expression of human papiloma virus( HPV)16 E6 gene in cervical cancer Siha cells. Methods The short hairpin RNA expression vector was transfected into cervical cancer Siha cell line with liposomes. The expression of E6 was detected with fluorescence quantitative RT-PCR and flow cytometry. Results The short hairpin RNA expression vector inhibited the expressions of E6 mRNA and protein. The inhibition ratio of E6 mRNA reduced to 92.65% and protein inhibition ratio reached to 84.40%. The braking effect lasted for more than 4 weeks. Conclusion The short hairpin RNA expression vector could specifically and effectively inhibit the expression of HPV16 E6 gene for a long time.
