摘要
目的探讨蜕皮甾酮对胰岛素抵抗HepG2细胞磷脂酰肌醇3激酶(PI3K)与葡萄糖转运蛋白-4(GLUT-4)蛋白表达的影响。方法胰岛素抵抗HepG2细胞模型建立后,培养液中加入蜕皮甾酮共同孵育,观察蜕皮甾酮及吡格列酮对模型细胞葡萄糖掺入率的影响;应用免疫细胞化学染色法及Westernblot等方法观察蜕皮甾酮对胰岛素抵抗HepG2细胞PI3K与GLUT-4蛋白表达的影响。结果与模型细胞组比较,1×10-5M蜕皮甾酮可使胰岛素抵抗HepG2细胞PI3K、GLUT-4蛋白的表达增加(P<0.05)。结论蜕皮甾酮的胰岛素增敏作用可能与胰岛素信号转导分子PI3K、GLUT-4蛋白的表达增强有关。
Objective To investigate the effect of ecdysterone on protein expressions of phosphatidylinositol 3-kinase(PI3K) and glucose transporter 4(GLUT-4) in the insulin-resistant(IR) HepG2 cells. Methods After establishment of the insulin resistant HepG2 cell model induced by high concentration of insulin. The HepG2 cells were incubated with 1×10^-5 M ecdysterone for 24 h. lmmunocytochemical assay was used to assess the expression of PI3K and GLUT-4 was determined by Western blotting. Results The incubation of insulin resistance cells with ecdysterone 1×10^-5M could significantly increase the protein expressions of PI3K and GLUT-4 in the IR HepG2 cells (P〈0.05). Conclusion Ecdysterone could improve the insulin sensitivity in IR HepG2 cells probably by uprcgulating the expression of insulin signaling proteins such as PI3K and GLUT-4.
