摘要
目的为了完善小鼠早期胚胎体外培养体系,探讨抗氧化剂维生素E、维生素C及槲皮素对体外授精胚胎发育的影响。方法以CZB为基础培养液,添加100μmol/L维生素E、200μmol/L维生素C和0.5μmol/L槲皮素作为实验组,对比各组胚胎发育到四细胞至囊胚各个不同阶段的发育率。结果实验组二细胞-四细胞发育率都高于对照组,其中以添加0.5μmol/L槲皮素组发育率为最高,然而维生素E、维生素C及槲皮素各组之间差异不显著。同样实验组二细胞-八细胞发育率也高于对照,以添加100μmol/L维生素E组发育率为最高,与其他各组相比,差异也无显著性。实验组二细胞-囊胚发育率高于对照,以添加200μmol/L维生素C组发育到囊胚率最高,维生素E、维生素C及槲皮素各组之间相比,差异极显著。故以添加200μmol/L维生素C对胚胎发育有明显的促进作用。结论3种抗氧化剂中,以200μmol/L维生素C对体外受精胚胎的促进作用最为明显,能得到较高的囊胚率。
Objective To improve mouse early embryo culture system and investigate the effects of vitamin E, vitamin C and quercetin on the development of mouse embryo in vitro fertilization. Methods The CZB was used as basic culture medium, added with 100μmol/L vitamin E, 200μmol/L vitamin C and 0.5μmol/L quercetin respectively as experiment groups. The cultured ceils were observed by light microscope to investigate the embryo development rate in the different phase from two-cells to blastocyst. Results Embryo development rates of two to four cells are higher in experiment groups than in control group, supplementation of media with quercetin at the concentration of 0.5 μmol/L is the highest among experiment groups, but with no differences between experiment groups. Embryo development rates of two to eight cells are higher in experiment groups than in control group, supplementation of media with vitamin E at the concentration of 100μmol/L is the highest among experiment groups, with no differences between experimental groups. Embryo development rates of two to blastocyst are higher in experiment groups than in control group, supplementation of media with vitamin C at the concentration of 200μmol/L is the highest among experiment groups, with significant differences between experimental groups.. So supplementation of media with vitamin C at the concentration of 200μmol/L can significantly improve embryo development. Conclusion Vitamin C at the concentration of 200μmol/L promotes the development of in vitro fertilizated mouse embryo among three kinds of antioxidants, has relative high blastocyst rates.
