IFN-α2b体外对人眼葡萄膜黑色素瘤细胞端粒酶活性的影响 被引量:1

摘要 目的:研究端粒酶抑制剂IFN-α2b对人葡萄膜黑色素瘤细胞端粒酶活性的抑制作用。方法:采用原代培养的人葡萄膜黑色素瘤细胞,用特定时间下不同浓度以及特定浓度下不同时间的端粒酶抑制剂IFN-α2b作用于体外培养的黑色素瘤细胞,并设立空白对照组。采用PCR-ELISA法及PCR-PAGE法测定细胞中端粒酶活性的变化,采用单因素方差进行统计分析。结果:随着IFN-α2b浓度的增加及作用时间的延长,端粒酶的活性出现逐步的下降,在药物浓度达到50kU/L以及作用时间达到24h后,出现明显的抑制作用,并在500kU/L及48h达到抑制高峰。结论:IFN-α2b可有效降低体外培养的人葡萄膜黑色素瘤细胞的端粒酶活性,并呈浓度和时间依赖性。 AIM: To study the restrain effects of telomerase inhibitor IFN-α 2b ( Interferon-α 2b) on the telomerase activity to uveal melanoma cells of human eyes. METHODS: Using IFN-α 2b individually with different concentration and time acted on the primary cultured uveal melanoma cells of human eyes and compared with the controls. The changes of telomerase activity were observed by PCR-ELISA and PCR-PAGE. The restrain results were compared with a one-way analysis of variance ( ANOVA). RESULTS. The tolemerase activity depressed as the increasing concentration and time of IFN-α 2b. Significant inhibition appear at the concentration attained 50kU/L or the time attained 24 hours. At the point which 500 kU/L or the time attained 48 hours the tolemerase activity depressed very obviously. CONCLUSION: As an effective telomerase inhibitor,IFN-α 2b can depress the tolemerase activity of uveal melanoma cells effectively. And it appeared concentration and time dependency.
