摘要
疫苗是捻转血矛线虫病防治研究中的热点之一,已有多种疫苗候选抗原得到鉴定,多数抗原在线虫的肠道表达,其天然提取物能够提供有效的免疫保护,减卵率可达70%~95%.由于糖基化作用、空间构象的错误折叠等因素的影响,疫苗候选抗原的重组形式不能为动物提供有效的免疫保护;免疫策略难以制定、佐剂难以选择也成为疫苗研制中的桎梏.因此,开发与天然提取物具有相似功能的转基因蛋白成为目前疫苗研究中的焦点,秀丽隐杆线虫表达系统和寄生性线虫细胞系的培育是研究中比较有潜力的两个方向;同时,RNAi技术等新方法的运用也加快了新的疫苗候选抗原的鉴定进程.
Substantial progress has been made in identifying several antigens from Haemonchus contortus, mostly expressed on the surface of the worm gut which are exposed to the blood meal. Their native form can stimulate effectively protective immunity with the reductions of 70%-95% in fecal egg output. Due to the influence of glyeosylation and conformational epitopes, little, even no utility, affects against Haemonchus contortus using their recombinant form of these antigens on goats. Undefined vaccine strategies and unsuitable adjuvant are also the hurdles blocking the development of vaccine. Now, much more regards focus on the exploring transgene proteins with similar functions to the native forms by inspiring means, such as expression in Caenorhabditis elegans or culture of parasitic cell lines. This review focuses on the advance on exploration of vaccine candidates from Haemonchus contortus, analyzing the limitation in vaccination using recombinant antigens, and makes a prediction of the trends on parasitic vaccine.
