优化复方CK制剂对肾性高血压大鼠的药效学

摘要 通过动物实验进行优化复方CK的药效学研究。取雄性wistar大鼠80只,随机分为假手术组(n=12)和模型组(n=68)。采用二肾一夹法复制肾性高血压大鼠模型。取造模成功的大鼠随机分为五组:模型组,优化复方CK高、中、低剂量组,卡托普利组,后4组灌胃给予相应的药物,模型组和假手术组灌服蒸馏水,每日1次,连续灌胃8周。根据体重随时调整给药量。观察药物对模型动物收缩压及血浆肾素活性、血管紧张素II和一氧化氮含量的影响。给药8周后,与模型组相比,优化复方CK高、中剂量组能显著地(P<0.01)降低模型动物收缩压、血浆肾素活性和血管紧张素II含量,升高血浆一氧化氮含量。由此可知优化复方CK有降压作用,其作用可能与降低血浆肾素活性和血管紧张素II含量、促进血液中的一氧化氮合成与释放有关。 To study the phannacodynamics of the grading-up compound r-ecipe CK by animal experiments. Eighty male Wistar rats were randomly divided into sham group( n = 12) and model group ( n = 68). Male Wistar rats of model group were made to be renal hypertension by "2KIC" and then randomly divided into five groups:model group,high-dose,middle-dose and low-dose grading-up compound recipe CK group, and captopril group, and then posterior four groups were given drugs accordingly by intragastric administration, model group and sham group were given Distilled Water by intragastric administration, once daily, for eight weeks continuously, and the volumes of the drug were adjusted any time according to the changes of their weight. The systolic blood pressure of the rats, plasma renin activity and content of Angiotensin Ⅱ and NO was measured to observe the effect of grading-up compound recipe CK on the model animals. After eight-week treatment, compared with model group, the high-dose and middle-dose grading-up compound recipe CK groups could remarkably ( P 〈 0.01)reduce the systolic blood pressure,plasma renin activity and content of Angiotensin Ⅱ of the model animals, and step up plasma content of NO of the model animals. The grading-up compound recipe CK can reduce renal hypertension, the effect is probably concerned with reducing plasma renin activity and content of Angiotensin Ⅱ and encouraging the composition and release of blood NO.
