平喘方对支气管哮喘模型小鼠气道重建的影响 被引量:10

摘要 目的:通过研究平喘方对支气管哮喘模型小鼠外周血和支气管肺泡灌洗液(bronchoalveo larlavage fluid,BALF)巨噬细胞炎性蛋白-1α(macrophage inflammatory protein-1α,MIP-1α)和免疫球蛋白E(immuno-globulin E,IgE)含量及CD86分子表达的影响,探索平喘方对小鼠支气管哮喘模型气道重建的作用机制。方法:采用卵清白蛋白腹腔注射和雾化吸入致敏,建立BALB/c小鼠支气管哮喘模型。50只雄性BALB/c小鼠随机分为空白对照组、哮喘模型组、地塞米松组和平喘方小剂量、常规剂量组,每组10只。造模后4周,予相应药物灌胃治疗8周。用亲和素-生物素复合物酶联免疫吸附测定法检测外周血和BALF中MIP-1α水平;用酶联免疫吸附测定法检测血清和BALF中IgE水平;直接免疫荧光流式细胞术检测外周血和BALF中CD86+、CD3-CD86+细胞百分率;利用普通光学显微镜观察支气管哮喘模型小鼠肺组织病理学改变。结果:模型组外周血和BALF中MIP-1α、IgE含量及CD86+、CD3-CD86+细胞百分率均明显高于空白对照组(P<0.01);治疗8周后,各治疗组外周血和BALF中MIP-1α、IgE含量和CD86+、CD3-CD86+细胞百分率均明显低于模型组(P<0.01);各治疗组之间两两比较,外周血和BALF中MIP-1α和IgE含量差异无统计学意义(P>0.05),平喘方小剂量组外周血中CD3-CD86+细胞百分率高于地塞米松组(P<0.05)。肺组织病理显示,各治疗组细支气管壁上胶原纤维较模型组明显减少。结论:平喘方能降低支气管哮喘模型小鼠MIP-1α和IgE水平,下调CD86分子表达,这可能是平喘方减轻支气管哮喘模型小鼠的症状,改善支气管哮喘气道炎症的作用机制之一。 Objective: To study the effects of Pingchuan Recipe, a compound traditional Chinese herbal medicine for treating bronchial asthma, on macrophage inflammatory protein-1α (MIP-1α) and immunoglobulin E (IgE) contents and CD86 expression in a mouse model of bronchial asthma, and to investigate the mechanism of Pingchuan Recipe in regulating airway remodeling in mice with bronchial asthma. Methods: Fifty BALB/c mice were randomly divided into normal control group, untreated group, dexamethasone group, low-dose Pingchuan Recipe group and normal-dose Pingchuan Recipe group. Bronchial asthma in mice was induced by intra-abdominal injection of mixed ovalbumin and stimulation by inhaling 5% ovalbumin. Mice in the normal control group and the untreated group did not receive treatment, but mice in the other groups were administered intragastrically with 50 g/(kg · d) Pingchuan Recipe, 25 g/(kg · d) Pingchuan Recipe and 0.75 mg/(kg · d) dexamethasone, respectively. After consecutive treatment for 8 weeks, the MIP-1α contents in peripheral blood and bronchoalveolar lavage fluid (BALF) were measured by an avidin biotin complex-enzyme-linked immunosorbent assay (ABC-ELISA) system, and the IgE contents in serum and BALF were measured by ELISA method. Direct immunfluorescence-flow cytometry was used to detect the percentages of CD86+ and CD3-CD86+ cells in peripheral blood and BALF. General histological changes of the lung tissue were observed by HE staining and collagen fiber staining 12 weeks after activation. Results: Compared with the normal control group, the contents of MIP-1α, the percentages of CD86+ cells and CD3-CD86+cells in peripheral blood and BALF, and the contents of IgE in serum and BALF were raised significantly (P〈0.01). The contents of MIP-1α, the percentages of CD86+ cells and CD3-CD86+ cells in peripheral blood and BALF, and the contents of IgE in serum and BALF in the treated groups were lower than those in the untreated group (P〈0. 01). The contents of MIP-1α in peripheral blood and BALF and the contents of IgE in serum and BALF had no significant differences among the treated groups (P〈0.05). The percentage of CD3-CD86+ ceils in peripheral blood in low-dose Pingchuan Recipe group was lower than that in the dexamethasone group. Compared with the untreated group, the collagen deposition in the wall of bronchiole in the treated groups was decreased. Conclusion: Pingchuan Recipe can decrease the contents of MIP-1α, IgE and the expressions of CD86 in peripheral blood and BALF, which may be one of its mechanisms in improving chronic airway inflammation and inhibiting airway remodeling.
