摘要
从枸骨叶中提取总黄酮并测定其含量。通过超声辅助提取,以芦丁为对照品,通过紫外—分光光度法测定总黄酮含量。在单因素试验基础上采用L9(34)正交设计,在提取温度为70℃、料液比为1∶12(W∶V)的40%乙醇中超声提取25min的条件下,枸骨叶总黄酮提取率最高达4.413%,提取级数为2级,重复性试验的相对标准偏差(RSD)为2.54%(n=5)。测定方法简便、快速、费用低,运用超声技术提取总黄酮效果较好。
The total flavonoids were extracted and determined, which were from llex comuta Lindl.. The total flavonoids in the llex cornuta Lindl. were extracted with ultrasonic-assisted technology, and determined through ultraviolet spectrophotometry by using rutin as standard sample. The effect factors were studied through orthognnal experiments L9 ( 3^4 ) according to the single factor experiment. The optimal extraction conditions can be gotten as follows: ratio of material to solvent in 60% ethanol = 1 : 12 ( W : V) , temperature 70 ℃, the time 25 min, extracting time twice, RSD 2.54% (n = 5) and the extracting ratio 4. 413%. The method to determine total flavonoids was simple, rapid and low cost. The results show that it is better by using uhrasnnic technology to extract total flavonoids in llex cornuta Lindl.
