RP-HPLC determination of lansoprazole enterosoluble tablets in human plasma

目的:建立高效液相色谱法测定人血浆中兰索拉唑肠溶片的浓度。方法:采用KromailC18色谱柱(4.6mm×250mm,5μm),以甲醇-水(70∶45)为流动相,流速1.0mL·min-1,检测波长284nm。结果:兰索拉唑的线性范围为2.0～300.0ng.mL-1,最低定量限为2.0ng.mL-1,回归方程A=2.802×103C-8.224×103,r=0.9997(权重1/c2×106)。结论:该方法简便、快速、准确,适用于临床上兰索拉唑药代动力学的研究。

Objective：A convenient RP-HPLC method was developed for measuring the concentration of lansoprazole in human plasma.Method：Chromatography was carried out on a Kromail C18 column（4.6 mm×250 mm,5 μm）with methanol-water（70∶45）as mobile phase,at a flow rate of 1.0 mL·min-1 and the detection wavelength was set at 284 nm.Results：The linearity range of lansoprazole was 2.0-300.0 ng·mL-1,the lowest limit of quantification was 2.0 ng·mL-1,the regressive equation was A=2.802×103C-8.224×103,r=0.9997（the weight factor was 1/c2×106）.Conclusions：The method is shown to be convenient,accurate,rapid and suitable for determination of lansoprazole in human plasma and the studies on clinical pharmacokinetics.
