摘要
目的:建立固相萃取-反相高效液相色谱法测定人血浆中长春瑞滨的浓度。方法:样品经C18固相萃取小柱净化后,采用HPLC紫外检测法,色谱柱为HypersilODS柱(250mm×4.6mm,5μm),流动相为乙腈:0.05mol·L-1磷酸二氢钾溶液(68∶32,含0.1%三乙胺,磷酸调pH=4),流速为1.0mL·min-1,柱温为室温,检测波长268nm。结果:长春瑞滨在0.1~4.8μg.mL-1内呈良好的线性关系(r=0.9994),最低检测浓度为50ng.mL-1,方法回收率在96.8%~103.0%,萃取回收率在76.7%~83.0%。日内、日间RSD(n=5)均小于6.2%。结论:本方法简便、准确,可靠,可用于人体血浆中长春瑞滨浓度的测定。
Objective:To develop an SPE-HPLC method for the determination of vinorelbine in human plasma.Method:The sample was cleaned with an SPE-C18 cartridge,separated and quantitatively determined by reversed-phase HPLC with an ultraviolet detector.The analytical column was packed with Hypersil ODS(250 mm×4.6 nm,5 μm),the mobile phase was acetonitrile-potassium dihydrogen phosphate solution(68∶32,containing 0.1% triethyamine,adjusted to pH 4.0 by phosphoric acid),the flow rate was 1 mL·min-1,and detection wavelength was set at 268 nm.Results:The linear range was 0.1-4.8 μg·mL-1 for vinorelbine in human plasma.The correlation coefficients was 0.9994.The low limit of detection was 50 ng·mL-1.The method recoveries were 96.8%-103.0%.The extraction recoveries were 76.7%-83.0%.Within-day and between-day RSDs(n=5)were less than 6.2%.Conclusion:The method is simple,reliable and sensitive for determination of vinorelbine in human plasma.
