摘要
目的检测IL-18、IL-6、IL-10在糖尿病大鼠肾脏的表达,探讨其在糖尿病肾脏病变过程中的作用。方法将40只雄性Wistar大鼠随机分为正常对照4周组(NC1组)、8周组(NC2组)、糖尿病4周组(DMI组)、8周组(DM2组),每组10只。DM组给予一次性腹腔内注射60mg/kg链脲佐菌素建立糖尿病大鼠模型。检测各组大鼠体重、肾重、尿微量白蛋白排泄率(UAER)。用免疫组化方法检测肾组织IL-18、IL-6、IL-10的表达,利用计算机图像分析系统进行定量分析。结果DM组大鼠的肾重指数(KWI)、UAER较NC组显著增高;DM2组较DM1组明显增高;IL-18、IL-6在NC组肾组织中仅少量表达,而在DM1组表达明显增多,在DM2组表达增高则更为显著。IL-10在NC组丰富表达,而在DM1组表达明显减弱,在DM2组几乎无表达。肾脏局部IL-18、IL-6与IL-10的表达呈显著负相关。结论糖尿病大鼠肾脏局部IL-18、IL-6表达明显升高、IL-10水平显著降低,IL-18、IL-6与IL-10之间平衡的紊乱在糖尿病肾脏损害病程中发挥重要的作用。
Objective To investigate the potential role of IL-18,IL-6 and IL-10 expression in the pathogenesis of diabetic nephropathy in rats. Method Forty male Wistar rats were divided in random: normal control group of 4 weeks(NC1 group) and 8 weeks(NC2 group), diabetic group of 4 weeks (DM1 group) and 8 weeks(DM2 group) (n = 10 in each group). The rats in DM groups were induced to diabetic mellitus by intraperitoneal injection of streptozotocin(STZ, 60 mg/kg). The expression of IL-18, IL-6 and IL-10 in the renal tissue was detected by immunohistochemistry staining and computer image-pattern analysis system. At the same time kidney weight index(KWI) and UAER were measured. Result The KWI and UAER were significantly higher in DM groups than in NC groups(P〈 0. 01). The expression of ID18 and IL-6 was rather weak in NC groups,while much elevated in DM1 group(P〈0. 01), and strikingly high in DM2 group(P〈0. 01). In contrast, the expression of IL-10 was abundant in NC groups, while decreased in DM1 group(P〈0. 01 ) and fewer in DM2 group(P〈 0. 01). There was a significantly negative correlation between the expression of IL-18, IL-6 and IL-10. Conclusion The expression of IL-18 and IL-6 is enhanced,but that of IL-10 significantly reduced in the renal tissue of diabetic rats. The disorder of the balance between IL-18, IL-6 and IL-10 may play an important role in the progression of renal lesion in diabetic rats.
