摘要
甘蓝型油菜是由白菜型油菜(A组基因型)与甘蓝(C组基因型)杂交后自然加倍的异源四倍体芸薹属植物。其中含有A组与C组两种基因型的染色体,而FLC1与FLC3是A、C组中共同拥有的FLC基因。通过序列比对,分别寻找出A、C组基因中FLC1与FLC3基因的特异性SNP位点,并设计了3对SNP特异性引物对。采用特异性引物延伸法,分别对来自白菜型油菜、甘蓝以及甘蓝型油菜的不同材料进行检测。利用这3对特异性SNP引物对,成功地对油菜中A、C组的FLC1与FLC3基因进行了有效的分型,为研究甘蓝型油菜中A、C组来源的FLC1与FLC3表达奠定了基础。
Brassica napus is a kind of allotetraploid of Brassica genus, which is hybridized by B. Campestris L. (A genome) and B. oleracea L. (A genome). It contains two genotypic chromosomes from genome A and C, and these two genomes have FLC1 and FLC3 in common. Three specific primer pairs are designed by finding the specific SNP site respectively in FLC1 and FLC3 of genome A and C. This project was to detect the genome difference among B. Campestris, B. olerclcea and B. napus by PCR amplification tool with three pairs of specific primers for each DNA sample, and we successfully genotyped FLC1 and FLC3 genes in genome A and C of Brassica corps. This project was aimed to set the foundation for the further research on the expressing of FLCI and FLC3 genes originated from genome A and C in Brassica napus.
