真性红细胞增多症患者JAK2外显子12突变研究 被引量:1

摘要 JAK2获得性功能性突变的发现使人们对骨髓增殖性疾病(MPD)的认识有了突破性进展。真性红细胞增多症(PV)是一种克隆性的以红细胞异常增殖为主的MPD。JAK2V617F点突变位于外显子14。最近发现JAK2V617F点突变阴性的PV患者存在JAK2外显子12的突变,这些外显子12的突变改变了539、543及547位密码子附近的多种核苷酸,并且JAK2外显子12突变的位置均有微小的差别,与JAK2V617F一致的结构形成了鲜明的对比。我们应用PCR扩增12例JAK2V617F点突变阴性PV患者的JAK2外显子12片段,通过基因测序检测JAK2外显子12突变,并结合临床和实验室资料进行分析,探讨PV患者的JAK2外显子12突变的发生率及其临床意义。
