应用VEGF防治肢体严重创伤后骨质疏松的实验研究 被引量:3

摘要 目的研究局部应用血管内皮生长因子(vasular endothelial growth foutor,VEGF)防治肢体严重创伤后局部骨质疏松的作用,为临床应用提供实验依据。方法参照Allen氏垂直重量打击法(weight-dropping,WD法)制作新西兰家兔桡骨远端严重损伤动物模型。将实验动物随机分为实验组(局部注射pHVEGF165质粒)与对照组(局部应用生理盐水组),每组各15只。实验组于伤后即刻局部(桡骨远端周围)注射pHVEGF165质粒50 ug,对照组局部注射生理盐水。于给药后2、4、6、8周观察桡骨远端血流量及骨密度变化。治疗后8周处死动物,行病理学检查。结果实验组伤后2、4、6、8周血流量优于对照组,桡骨远端骨密度明显高于对照组(P<0.05)。实验组病理学检查见骨小梁增多、增粗。结论局部注射pHVEGF165质粒可以改善肢体严重创伤后局部血流量,预防局部骨质疏松的发生。 Objective To approach the effect of local application of VEGF for curing bone rarefaction after serious limbs crush injury,and to provide experimental basis for clinical treatment. Methods Animal model of New Zealand rabbit's limbs(distal radius) crush injury were produced with reference to Allen's vertical combat weight (weight-- dropping, WD method) and were randomly divided into experimental group (local injection with pHVEGF plasmid) and the control group (local injecting with normal sodium) ,and each group have 15 New Zealand rabbits. Experimental group were injected instantly with 50 ug pHVEGF plasmid after limbs crush injury,and control group were injected with normal sodium. The diversity of regional blood flow and bone density of distal radius were detected after 2,4, 6 and 8 weeks after local injection. Then,after 8 weeks ,the animals were sacrificed to make pathologic detection. Resuits Regional blood flow of experimental group was better than that of control group after 2,4,6 and 8 weeks,and bone density of distal radius was so ,and P〈0.05. There was a significant difference. Pathologic detection showed that bone trabecula of experimental group was increased and thickened more. Conclusion Local injection of pHVEGF plasmid can improve regional blood flow of crush injury limbs and prevent regional bone rarefaction.
