摘要 为了确定MSTN RNAi双元表达载体pEGFP-N1-M1-M2对成肌细胞增殖和分化的影响,利用脂质体介导法将该双元表达载体转染成肌细胞,MTT法检测MSTN RNAi双元表达载体对成肌细胞增殖的作用,Giemsa染色检测pEGFP-N1-M1-M2双元表达载体对成肌细胞融合率的影响。试验结果表明,pEGFP-N1-M1-M2双元表达载体有促进成肌细胞增殖和分化的作用,能够促进成肌细胞大量融合。该结果为进一步探讨MSTN基因的作用机理提供了更为便捷的研究方法。 To decide the effect of pEGFP-N1-M1-M2 binary vector to myoblast cell, the binary vector was transfected into myoblast ceils by lipidosome, and myoblast hyperplasia and differentiation was detected by MTT and Giemsa assay. The results showed that pEGFP-N1-M1-M2 binary vector could promote myoblast hyperplasia, differentiation and fusion.The results provided easier experimental way to study the mechanism of MSTN gene.
