摘要
应用尿激酶原(proUK)基因重组复制缺陷型腺病毒载体,防治家兔髂动脉内膜剥脱后新生内膜的增生。结果显示:导入proUK基因三周后,血管组织内proUK基因的mR-NA及其表达产物明显高于导入野生型腺病毒对照组;血管造影及切片显示血管狭窄程度较轻,新生内膜与中膜面积比(I/M)明显低于对照组。提示:血管内膜剥脱后导入proUK基因,通过对抗血管内膜损伤后的血栓形成和血小板的沉积,从而抑制由血栓及血小板促发的新生内膜过度增生,具有潜在的应用价值。
The replication dificient adenovirus vector containing proUK gene(Ad/proUK) was used for treatment of neointima proliferation of intima denudated iliac artery in rabbit. The results from angiography and observation of light microscopy showed that the narrowness of the vessels transfected with proUK gene were lighter compared to control group, and the neointimal area/medial area ratio(I/M) in experimental group was markedly lower. These findings reveal that proUK gene can suppress neointima proliferation following intima injury through preventing platelet deposition and thrombolysis. This may have a potential theraputic value.
