程序性死亡因子5在Graves病和桥本甲状腺炎中表达的初步研究 被引量:2

A PRELIMINARY STUDY ON EXPRESSION OF PDCD5 IN PATIENTS WITH GRAVES DISEASE AND HASHIMOTO'S THYROIDITIS

摘要 目的探讨促凋亡基因PDCD5 mRNA在初诊Graves病(GD)和桥本甲状腺炎(HT)患者外周血单个核细胞(PBMC)中的表达。方法半定量RT-PCR法测定50例GD和30例HT患者PBMC中PDCD5 mRNA表达的阳性率和表达水平。结果GD和HT患者PBMC中PDCD5 mRNA表达的阳性率和表达水平均高于正常对照组,其中HT患者PDCD5 mRNA表达的阳性率显著高于正常对照组(P<0.05),GD、HT、正常对照三组间PDCD5 mRNA表达水平差异有统计学意义(P<0.01),HT组PDCD5 mRNA表达水平显著高于GD组(P<0.01)。患者PDCD5 mRNA表达水平与甲状腺微粒体抗体(TM-Ab)、甲状腺球蛋白抗体(TG-Ab)呈显著正相关(P<0.01),与血清T4水平及|lgTSH|呈显著负相关(P<0.05)。结论PDCD5 mRNA在GD和HT患者PBMC中表达升高,可能与GD和HT淋巴细胞凋亡异常有关。 Objective To study the expression of PDCD5 in peripheral blood monocyte (PBMC) from normal subjects and patients with graves disease (GD) and hashimoto's thyroiditis (HT). Methods The expression of PDCD5 was assayed by semi - quantitative reverse Reverse transcription - Polymerase chain reaction (RT - PCR). Results Compared with normal group, the positive rates of PDCD5 were increase in GD group and HT group. Positive rate was significantly higher in HT group than that in normal group(P〈0.05). The relective expression level was also higher in GD group and HT group than that in normal group(P〈0.01). The relective expression level was higher in HT group than that in GD group(P〈0.01). A statistically significant correlation was found between PDCD5 and TM- Ab, TG- Ab, T4, | 1gTSH| (P〈0.05). Conclusion These results suggest that the higher expression of PDCD5 mRNA may be involved in abnormal apoptosis of lymphocytes in GD and HT patients.
