钙通道阻滞剂对重度冻僵大鼠血液有形成分的影响

摘要 目的观察维拉帕米(verapanil)对冻僵大鼠血液有形成分的影响。方法实验动物为健康雄性wistar大鼠,随机分为5组,为对照组、冻僵组、干预1组、干预2组和干预3组。对照组于室温20℃下饲养,干预1、2、3组冷暴露前30min分别腹腔注射维拉帕米5、10和20mg/kg,冻僵组和干预组大鼠-30℃环境进行冷暴露80min后复温4h。观察血液WBC、RBC、PLT数量和肛温的变化。结果冻僵组和干预1组WBC数量明显高于对照组(P<0.01),干预2组WBC数量高于对照组(P<0.05),干预3组WBC数量与对照组比较,差异无统计学意义(P>0.05);各组红细胞数量差异无统计学意义(P>0.05);冻僵组PLT数量较对照组显著增高(P<0.01),而干预组PLT数量与对照组比较,差异无统计学意义(P>0.05);冻僵组和干预组肛温均低于31℃,与对照组比较,温差差异P<0.05,干预3组温差明显低于冻僵组(P>0.05)。结论10mg/kg维拉帕米能有效调节重度冻僵大鼠的血液有形成分数量,改善血液循环,防止体温过低的副效应。 [ Objective] To observe the effects of verapamil on the blood visible component of the frozen stiff rats. [ Methods] Healthy male Wistar rats were randomly divided into five groups: control group, frozen stiff group, interventional group 1, interventional group 2 and interventional group 3. Control group was bred at the room temperature of 20 ℃ . Verapamil of 5, 10, 20 mg/kg were injected into the abdominal cavity of the rats of the Interventional groups 1, 2, 3 respectively. The rats of the frozen stiff and interventional groups were exposed to cold condition at the temperature of -30℃ for 80min and then rewarmed for 4 hours. The changes of the counts of WBC,RBC, PLT and the level of anus temperature were observed. [ Results ] The counts of the WBC of frozen stiff group and interventional group 1 were significantly higher than that of control group (P 〈0. 01 ) ; that of interventional group 2 was also higher than the control group (P 〈0. 05); the difference of WBC count between interventional group 3 and control group was not significant ( P 〉 0. 05 ) ; The counts of the RBC between each 2 groups were not significantly different ( P 〉 0. 05 ) ; The counts of the PLT of frozen stiff group was significantly higher than that of control group ( P 〈 0. 01 ), while that between the interventional groups and control group were no significant ( P 〉 0. 05 ). The anus temperature levels of the frozen stiff group and interventional groups were all lower than 31 ℃, compared with the control group temperature the difference was significant ( P 〈0. 05 ) ; the temperature difference of interventional groups 3 were significantly lower than that of frozen stiff group(P 〈0. 05). [ Conclusion] 10 mg/kg verapamil can effectively regulate blood visible component, improve blood circulation, and prevent the side effects of hypothermia in frozen stiff rats.
