冷冻去甘油红细胞制备的2种方法比较

摘要 目的改良冷冻去甘油红细胞的手工洗涤方法,以提高制品的质量。方法将70份红细胞悬液分成2组,每组35份,Ⅰ组为未改良三珠袋组,Ⅱ组为改良三珠袋组,2组均做红细胞甘油化处理于-80℃冰箱保存,经快速解冻后,做去甘油洗涤,对比2组洗涤后的红细胞制剂的红细胞回收率、游离血红蛋白、残余白细胞和血小板、体外溶血情况以及甘油残留含量。结果使用改良方法洗涤的红细胞的各项指标均较改良前有明显改善,且经t检验差异均有统计学意义(t值为8.64~24.51,均P<0.01)。结论改良的方法简单、有效,能提高产品质量。 [ Objective ] To reform the hand-made cleaning method of deglycerolizing frozen red cells to improve the quality of products. [ Methods] 70 samples of red cell suspension were divided into two groups, group I was unreformed three bead group and group II was the reform group; both the two groups of samples were treated by deglyeerolization and conserved in refrigerator under 80℃ ; then the samples were defrosted quickly and deglycerolized; comparison was made on the red blood cells recovery rate, free hemoglobin, residue white blood cell, blood platelet, vitro hemolysis and residual glycerine content. [ Results ] All indexes were significantly improved after the reform, the difference was significant ( t = 8.64 - 24.51 ; P 〈 0. 01 ). [ Conclusion ] The reformed method is simple and effective, which can improve the quality of products.
