摘要
目的:研究弓形虫感染后对雄性小鼠一氧化氮(nitric oxide,NO)水平的改变对睾丸生精细胞的影响。方法:选用9~10周龄雄性BALB/C小鼠60只,随机分为正常对照组、弓形虫速殖子感染组(0.5×103、1×103、2×103、4×103)。采用腹腔注射法,建立弓形虫急性感染模型,免疫组化SP法测定睾丸Bax、Bcl-2表达的变化,硝酸还原法测定睾丸NO含量。结果:感染小鼠睾丸组织的NO水平明显增高,睾丸细胞Bcl-2表达无明显改变,而生精细胞尤其是精母细胞的Bax表达明显增加。结论:弓形虫感染导致NO升高,并可能通过Bcl蛋白途径促使生精细胞,尤其是精母细胞的损伤。
Objective:To explore the effect of nitric oxide (NO) to germ cell after Toxoplasma gondii infecting. Methods:Sixty BALB/C mice (9-10 weeks) were randomly divided into five groups:negative control group, tachyzoites infected groups (0.5×10^3, 1 ×10^3,2×10^3,4×10^3). Recording the general state of mice. NO level in testis was determined by means of nitric acid reduction.To detecte the apoptosis of spermatogenic cell was used flow cytometry and the expression of Bax,Bcl-2 in testis. Resnits:The levels of NO in infected mice testis were increased ;the expression of Bcl-2 had no significant change, while Bax expression enhanced significantly,especially in spermatocytes.Conclusion:Toxoplasa gondii leads to the increase of NO,and may induce the injury of spermatogenic cell through Bcl protein family.
