阿司匹林对糖尿病大鼠脂肪细胞肿瘤坏死因子-α表达的影响 被引量:2

摘要 目的研究阿司匹林对糖尿病大鼠脂肪细胞肿瘤坏死因子-α(TNF-α)mR-NA表达的影响,为临床防治2型糖尿病及微血管并发症提供理论基础和实验依据。方法用高糖高脂饮食及腹腔注射小剂量链脲佐菌素方法建立2型糖尿病大鼠模型,分为模型组、阿司匹林组和西药对照组,另选正常大鼠作为正常组。阿司匹林组给予阿司匹林450 mg/(kg.d)灌胃,西药对照组给予二甲双胍134 mg/(kg.d)灌胃。给药1个月后检测血糖、血脂、血清胰岛素含量和脂肪细胞TNF-αmRNA表达的变化。结果模型组大鼠血糖、血脂、血清胰岛素含量明显上升,脂肪细胞TNF-αmRNA表达显著升高;阿司匹林组和西药对照组以上指标均显著降低,组间比较,阿司匹林组明显低于模型组和西药对照组(P均<0.05)。结论阿司匹林能有效降低2型糖尿病大鼠脂肪细胞TNF-αmRNA表达和血糖、血脂、血清胰岛素浓度,其作用机制可能是通过抑制炎症因子释放而改善了胰岛素抵抗状态,因此有可能成为治疗糖尿病并发症的新的有效药物。 Objective It is to explore the effect of aspirin on mRNA expression of lipoblast tumor necrosis factor- α(TNF- α ) in rats with type 2 diabetes mellitus(T2DM), so to provide experimental and theoretical evidence for clinical treatment for T2DM and capillary vessel complications. Methods The T2DM model of rat was established by feeding with diet of high energy and fat, and by giving low dose of streptozotocin(STZ) through intraperitoneal injection. All the models were randomly divided into model group, aspirin group and western medicine control group. Some healthy rats were chose as normal group. Aspirin group and metformin group were respectively given aspirin 450 mg/(kg·d) and metforrnin 134 mg/(kg· d) by intragastric administration. After one month' s treatment, blood sugar, blood fat, serum insulin and the mRNA expression of lipoblast TNF- α were measured and analyzed. Results The contents of blood sugar, blood fat, serum insulin obviously all increased, and the level of mRNA expression of lipoblast TNF- α significantly rose up in model group. But all the indexes above were significantly decreased in aspirin and western medicine treated group. Compared with each other, the level of every index of aspirin treated group was lower than that of western medicine treated group ( P 〈 0.05 ). Conclusion Aspirin can significantly lowed down the level of lipoblast TNF- α, blood sugar, blood fat and serum insulin in rats with T2DM. This mechanism may be that aspirin can improve the insulin resistance by inhibit the expression of inflammation factors, so it may be used as a effective drugs for treating complications of diabetes mellitus.
