寻常性银屑病外周血单一核细胞中T-bet和GATA-3的基因表达 被引量:1

Expression of T-bet and GATA-3 in Peripheral Blood Mononuclear Cells of Patients with Psoriasis Vulgaris

摘要 目的观察寻常性银屑病患者外周血单一核细胞(PBMCs)中Th1细胞转录因子T-bet和Th2细胞转录因子GATA-3的基因表达,探讨T-bet和GATA-3在寻常性银屑病发病机制中的意义。方法采用双抗体夹心法(ELISA)检测寻常性银屑病患者血清中Th1型细胞因子γ-干扰素(IFN-γ)和Th2型细胞因子白介素(IL)-4的水平;逆转录聚合酶链反应(RT-PCR)法分析PBMCs中转录因子T-bet和GATA-3的基因表达。结果寻常性银屑病患者血清中IFN-γ的水平明显高于正常对照组,IL-4的水平明显低于正常对照组(P均<0.05);患者PBMCs中转录因子T-bet的表达明显高于正常对照组,GATA-3的表达明显低于正常对照组(P均<0.05);患者血清IFN-γ浓度与T-bet表达水平呈明显正相关(r=0.803,P<0.05),与GATA-3的表达水平无相关性(r=-0.5244,P>0.05);血清IL-4浓度与T-bet和GATA-3的表达水平均无显著相关性(r分别为-0.072,0.452,P均>0.05)。结论Th1特异性转录因子T-bet可能通过调节Th1型细胞因子IFN-γ的产生,介导寻常性银屑病的发生和发展。 Objective To investigate the role of Thelper 1 (Th1) specific transcription factor T-bet and Th2 specific transcription factor GATA-3 in the pathogenesis of psoriasis, and to observe the gene expression of T-bet and GATA-3 in peripheral blood mononuelear cells (PBMCs) of patients with psoriasis vulgaris. Methods Enzyme-linked immunoabsorbent assay(ELISA) was used to detect the secretion of Th1 cytokine interferon(IFN-γ) and Th2 cytokine interleukin (IL-4) in the serum of psoriasis patients and health eontrols. The gene expression of T-bet and GATA-3 by PBMCs of psoriasis patients and health controls was analysed by reverse transcriptase-polymerase chain reaction (RT-PCR). Results Compared with health controls, IFN-γ concentration in the serum was markedly higher in psoriasis patients (P〈0.05), while IL-4 eoneentration was markedly lower (P〈0.05). The expression of T-bet by PBMCs of psoriasis patients was significantly higher than that in health controls (P〈0.05), but GATA-3 was significantly lower (P〈0.05). Serum IFN-γ concentration showed a significant eorrelation with gene expression of T-bet (P〈0.05). Conclusion Th 1 specific transcription factor T-bet might play an important role in the development and maintenance of psoriasis by a possible feedback loop together with IFN-γ.
