抗旱、耐盐基因P5CS转化向日葵自交系 被引量:3

摘要 以向日葵保持系75-33B和恢复系Ht80-97的子叶为转化受体材料,用携带P5CS基因的双元载体系统的根癌农杆菌进行叶盘转化,获得抗Kan的转化芽。提取转化芽总DNA进行PCR检测及PCR-Southern杂交检测,证实获得阳性抗性芽75-33B 2株,Ht80-97 4株,转化率分别为3.17%和4.65%。 The leaf discs of sunflower maintainer fine 75-33B and restorer fine Ht80-97 were transformed with PSCS gene mediated by Agrobacterium tumefaciens which carried a binary vector pCHF3/P5CS. Transgenic shoots were obtained and detected by PCR and PCR-Southem blotting. The results indicated that two transgenic shoots from maintainer line 75-33B and four from restorer line Ht80-97 were positive, and the frequency of genetic transformation was 3.17% and 4.65% respectively.
