姜黄素对宫颈癌细胞系体外增殖的影响及机制研究 被引量:1

摘要 目的探讨姜黄素对宫颈癌细胞系SiHa细胞体外增殖的影响及其对细胞周期的作用。方法10~60μmol/L姜黄素分别处理SiHa细胞后,苔盼蓝染色和四甲基偶氮唑蓝法检测癌细胞的生长活性,流式细胞仪分析细胞周期各时相的变化。结果不同浓度的姜黄素对宫颈癌细胞系SiHa细胞的增殖均有抑制作用,并呈剂量依赖性。药物处理后的细胞G1期的比例增加,S期减少,G2/M期细胞比例相对增多。与对照组比较,不同浓度姜黄素(10、20、40、60μmol/L)处理后,G2/M期比例分别为8.7%±2.1%、10.4%±1.3%、10.9%±2.2%、11.8±1.4%。结论姜黄素能抑制宫颈癌细胞的体外增殖,其机制可能与细胞周期阻滞有关。 Objective To investigate inhibitory effects of eurcumin on in vitro proliferation of human cervical carcinoma cel1 line and its possible mechanisms. Methods SiHa cell line was treated with different concentrations ( 10 -60μmol/L) of curcumin. MTT assay and trypan-blue staining were used to assess proliferation of SiHa cells line. Flow cytometry was used to detect changes in cell cycle. Results Different concentrations of curcumin ( 10 - 60μmol/L) exerted inhibitory effects on the SiHa cells line and shown a dose-dependent relation. Furthermore, flow cytometry showed that after treatment the number of ceils in G1 phase increased and the number of cells in S phase decreased , the number of cells in G2/M phases relatively increased. Compared with control group, the cell percentages in G2/M phase were 8.7% ± 2.1%, 10.4%± 1.3%, 10.9% ±2.2% and 11.8% ± 1.4% respectively after treatment with different concentrations of curcumin ( 10, 20, 40, 60μmol/L). Conclusion Curcumin could inhibit in vitro proliferation of the SiHa cells, and its mechanism may be related with cell cycle arrest.
