通心络抑制ET-1诱发的冠状动脉痉挛作用机制探讨 被引量:7

摘要 随机将日本大耳白兔分为对照组、通心络超微粉剂低、中、高剂量组及法苏地尔组。内皮素-1(ET-1)诱发冠状动脉痉挛(CAS)。监测兔体表心电图,记录Ⅱ导联T波变化,检测6 h后血清心肌钙蛋白I(cTnI)水平,RT-PCR法检测冠状动脉平滑肌Rho-kinase、PKC-δ、NF-κB的表达。结果高剂量通心络、法苏地尔组均可均可见明显降低T波增高幅度及cTnI的释放水平,5 min时大白兔冠状动脉平滑肌Rho-kinase、PKC-δ、NF-κB mRNA的表达水平明显低于对照组,两组间比较无统计学差异(P>0.05)。认为通心络可通过阻断Rho-kinase途径和PKC-δ途径抑制ET-1诱发的CAS。
