摘要 目的:研究凋亡抑制基因Survivin、血管内皮生长因子(vascular endothelial groth factor,VEGF)、p53蛋白在乳腺癌组织中的表达,并探讨其临床意义。方法:用免疫组织化学SP法,对60例乳腺癌组织(A组)、22例乳腺良性增生组织(B组)及10例乳腺癌旁正常组织(C组)中的Survivin、VEGF及p53蛋白进行检测。结果:Survivin阳性表达率在A组和B组分别为76.7%(46/60)和18.2%(4/22),在C组不表达(P<0.05);Survivin表达与乳腺癌病理组织学分级、HER2表达有关。VEGF阳性表达率在A组和B组分别为73.3%(44/60)和31.8%(7/22),在C组为10%(1/10)(P<0.05);VEGF表达与乳腺癌淋巴结转移、病理组织学分级、HER2表达及临床分期有关。p53阳性表达率在A组和B组分别为51.7%(31/60)和13.6%(3/22),在C组不表达(P<0.05);p53表达与乳腺癌肿瘤大小、淋巴结转移、病理组织学分级、HER2表达及临床分期均有关。在乳腺癌组织中Survivin的表达与VEGF、p53的表达呈正相关。结论:Survivin、VEGF及p53在A组的表达明显高于B组和C组,并与转移和病理分级等生物学特征有关,在乳腺癌发生、发展中可能起重要作用,Survivin有望成为乳腺癌基因治疗的新靶点。 Objective:To study the expression of Survivin,VEGF,p53 and their correlation in breast cancer. Methods : The expression of Survivin,VEGF and p53 was evaluated in 60 cases of breast cancer, 22 cases of hyperplasia of mammary gland and 10 cases of paracainoma normal tissues by immunohistochemical technique (SP). Results: The positive rates of Survivin were 76.7%,18.2% and 0% in breast cancer, hyperplasia of mammary gland and paracacinoma normal tissues (P〈0.05).The expression of Survivin was significantly correlated with the histological grade of breast cancer and HER2 in breast cancer. The positive rates of VEGF were 73.3% ,31.8% ,10% in breast cancer, hyperplasia of mammary gland and paracacinoma normal tissues, respectively (P〈0. 05).The expression of VEGF was significantly correlated with lymphonodes mestastasis, the histological grade of breast cancer, HER2 and clinical staging in breast cancer. The positive rates of P53 were 51.7%,13.6% and 0% in breast cancer, hyperplasia of mammary gland and paracacinoma normal tissues,respectively (P〈0.05).The expression of P53 was significantly correlated with tumor size, lymphonodes metastasis, the histological grade of breast cancer, HER2 and clinical staging in breast cancer. Conclusion: The expression of Survivin,VEGF,p53 in breast cancer is significantly higher than that in hyperplasia of mammary gland and paracacinoma normal tissues ,and correlated with metastasis, the histological grade of breast cancer.
