白介素-1β外膜介导小型猪冠状动脉粥样硬化病变的研究 被引量:8

摘要 目的:探讨炎症因子白介素(IL)-1β外膜介导小型猪冠状动脉粥样硬化病变及其可能机制。方法:小型雄性家猪16只随机分为2组,开胸手术分离冠状动脉左前降支和回旋支近端,对照组(n=8)血管外膜包裹吸附含生理盐水琼脂糖微粒悬液的纸巾;模型组(n=8)包裹吸附含IL-1β2.5μg琼脂糖微粒悬液的纸巾。2周后,冠状动脉造影观察管腔狭窄程度,光镜观察管腔病理学改变。结果:冠状动脉外膜包裹IL-1β血管段发生管腔狭窄,光镜可见病变血管段内膜增殖和炎症细胞聚集现象,外膜可见大量炎症细胞浸润。逆转录聚合酶链反应示模型组Rho激酶、单核细胞趋化蛋白-1、细胞间黏附分子-1和血管细胞间黏附分子-1mRNA表达强度高于对照组(P<0.05)。结论:IL-1β包裹冠状动脉外膜介导冠状动脉炎症反应及粥样硬化病变,上调细胞因子和黏附因子的表达是其主要作用途径之一。 Objective: To investigate the possible mechanism of coronary atberosclerosis induced by IL-1β through external membrane in mini-swine. Methods: Sixteen male mini-swine were randomly divided into model group (n=8)and control group (n =8). Chest operation was done by isolation of the left-anterior descending branch and circumflex branch proximaal part. Coronary artery membranna externa in swine was packed by paper towel absorbing saline agarose particle suspension in control group. But in model group coronary artery membranna externa was packed by paper towel absorbing IL- 1β (2.5 μg)agarose particle suspension. After 2 weeks, lumen narrow was observed by coronary arteriography. The vascular section in the treated site was taken out to be examined by pathology. The expressions of Rho kinase,MCP-1 ,VCAM-1 and ICAM-1 mRNA were measured by reverse transcriptionpolymerase chain reaction analysis (RT-PCR). Results: In the model group, luminal lessen, inner membrane proliferation and inflammatory cell aggregation were observed in the sections of which the external membrane were wrapped by IL-1β. A great number of inflammatory cell infiltrations were also observed on the external membrane. The expression levels of the Rho-kinase, MCP-1,VCAM-1 and ICAM-1 mRNA were obviously increased in model group compared with those in control group (P 〈 0.05). Conclusion: Coronary artery inflammatory reaction and atherosclerosis were induced by IL-1β wrapped around the external membrane. One of the most important ways was the upgrade of expressions of cytokines and VCAM-1 and ICAM-1.
