摘要
探讨白细胞介素18(IL-18)对人肾小管上皮细胞E-cadherin表达的影响,进一步明确IL-18促进肾小管上皮细胞转分化的机制。应用体外细胞培养技术培养人肾小管上皮细胞株(HK-2细胞)。采用终浓度分别为0、1、10、100 ng/ml的IL-18处理HK-2细胞24 h、48 h、72 h。然后应用免疫细胞化学法检测HK-2细胞E-cadherin的表达百分数;采用RT-PCR法检测HK-2细胞E-cadherin mRNA的表达。结果表明,IL-18呈剂量时间依赖性地降低E-cadherin阳性的HK-2细胞百分数(P<0.01),且呈剂量时间依赖性地减少HK-2细胞E-cadherin mRNA的表达(P<0.01)。提示IL-18可抑制肾小管上皮细胞E-cadherin的表达,促进肾小管上皮细胞转分化。
To investigate the effect of IL-18 on E-cadherin expression in renal tubular epithelial cells(TECs). Human proximal tubular epithelial cell line(HK 2) was cultured in vitro, and TECs were exposed to different concentrations (0, 1, 10, 100 ng/ml) of IL-18 for 24, 48 and 72 hours. At the end of each incubation, the expressions of the E-eadherin mRNA were assessed by RT-PCR, and the rate of E cadherin positive TECs was assessed by immunocytoehemistry. It was demonstrated that the rate of E-cadherin positive TECs was decreased in a dose- and time dependent manner when HK-2 cells were exposed to IL-18(P〈0.01). The expressions of E-cadherin mRNA were also decreased in a dose- and time-dependent manner when TECs were exposed to IL-18 (P〈0.01). It is concluded that IL-18 can promotes the epithelial-mesenchymal transdifferentiation by decreasing the expression of E-cadherin in TECs.
