槲皮素对乳腺癌细胞侵袭及其cripto表达的影响 被引量:1

摘要 目的观察槲皮素(Que)对乳腺癌细胞侵袭能力的影响,并探讨其可能机制。方法采用不同浓度的Que作用乳腺癌MDA-MB-468细胞,以软琼脂集落培养试验检测癌细胞集落形成数,以Boyden小室模型检测癌细胞侵袭力,分别以荧光实时定量RT-PCR和Western blot方法检测癌细胞cripto mRNA及其蛋白。结果经Que作用后,MDA-MB-468细胞恶性增殖和侵袭力均明显下降,且呈浓度依赖性(P均<0.05),其cripto mRNA和蛋白表达均明显下调,也呈时间和浓度依赖性(P均<0.05)。结论Que可明显抑制乳腺癌MDA-MB-468细胞的侵袭力,其机制可能与下调该细胞cripto基因表达有关。 Objective To study the effects and mechanism of quercetin(Que) on human breast cancer cell.Methods Human breast cancer MDA-MB-468 cells were treated with different doses of Que, the growth of anchorage independence of cancer cell was studied by colony formation in soft agar, and invasion ability were determined by Boyden chamber, the mRNA and protein of cripto were detected by real time RT-PCR and Western blot assay, respectively. Results The invasion ability and anchorage of MDA-MB-468 cells were decresed by Que treated and in a dose-dependent manner( P 〈 0.05) .The cripto mRNA and protein of the cells wese decresed and in a dose-dependent manner( P 〈 0.05). Conclusion Que can inhibit invasion of cell line MDA-MB- 468 through down-regulating of cripto expression.
