TAM诱导MA782细胞凋亡及其对Cyclin D1、p21/CIP1表达的影响

摘要 将TAM体外作用于MA782细胞。采用MTT法检测细胞增殖活性;采用流式细胞仪检测细胞凋亡和细胞周期分布;采用免疫组化ABC法检测细胞Cyclin D1、p21蛋白表达,并用病理图像分析软件进行半定量分析;采用RT-PCR法检测MA782细胞Cyclin D1 mRNA表达情况。结果显示TAM在体外能明显抑制MA782细胞生长,诱导细胞凋亡。TAM诱导后MA782细胞Cyclin D1 mRNA及蛋白表达下降,而p21蛋白表达上升(P均<0.01)。认为TAM诱导MA782细胞凋亡的机制可能与其下调Cyclin D1蛋白和mRNA表达及上调p21蛋白表达有关。
