摘要
目的探讨二氧化硅(SiO2)参与矽肺纤维化形成的机制。方法收集矽肺患者肺泡灌洗液中的巨噬细胞(AM),在体外以SiO2(50μg/ml)和不加血清的DMEM培养2、6、12、18、24、36 h,应用免疫细胞化学法检测AM中转化生长因子-β1(TGF-β1)水平;然后取培养18 h的AM上清与人肺成纤维细胞(FB)共同孵育6、12、18、24、36、48 h,同法检测FB中TGF-β1水平。结果经SiO2刺激的矽肺患者AM中TGF-β1表达增加(P<0.05);AM上清也可使FB中TGF-β1表达上调,经SiO2刺激的AM上清作用更明显(P<0.01)。结论SiO2可促进矽肺纤维化形成,其机制可能为上调AM和FB中TGF-β表达。
Objective To investigate the role of SiO2 in the pathogenesis of silicosis. Methods Human alveolar macrophages(AM) from bronchoalveolar lavage of silicosis patient were collected and exposed to SiO2 (50 μg/ml) for dif- ferent time (2,6,12,18,24,36 h ), then the conditioned supernatant were collected. The human lung fibroblasts (FB) were isolated by trypsin dispersion method, and incubated with the conditioned supernatant of AM for 6,12,18,24,36,48 h. Immunocytochemical method was used to detect the expression of TGF-β1 in AM and FB. Results The expression of TGF-β1 in AM exposed to SiO2 in vitro Was up-regulated( P 〈 0.05 ). Expression of TGF-β1 in FB incubated with the supernatant of AM increased(P 〈0.01 ), while the expression of TGF-β1 was higher in the FB incubated with the supematant of AM exposed to SiO2. Conclusion SiO2 can induce up-expression of TGF-β in AM and FB, which contribute to the pathogenesis of silicosis.
