O-2A祖细胞移植对大鼠脊髓损伤后运动功能恢复的作用 被引量:1

Effects of the Oligodendrocyte-type 2 Astrocyte (O-2A) Progenitor Cells Transplantation on Repair of Rat Spinal Cord Injury

摘要 目的研究O-2A祖细胞移植对大鼠脊髓损伤(SCI)后运动功能恢复的作用。方法从新生1~2 d SD大鼠大脑皮质中分离纯化O-2A祖细胞。80只SD成年大鼠,随机分为4组:A组为正常组,与其他3组同期饲养;其他3组均行T9脊髓左半侧离断手术,B组损伤处植入吸附生理盐水的明胶海绵(损伤对照组);C组损伤处移植50%O-2A祖细胞和50%1型星形胶质细胞(T1A)混悬液(O-2A+T1A组);D组损伤处植入含有纯化O-2A祖细胞的细胞悬液(O-2A移植组)。移植术后3、7、14、21、28 d分别以CBS及BBB方法进行大鼠神经功能评定。结果A组大鼠后肢CBS和BBB得分分别为100和21。O-2A祖细胞移植术后3、7、14、21、28 d,B、C、D组动物后肢神经功能均有不同程度的恢复,左后肢得分均显著低于右侧,差异均有高度统计学意义(均P<0.01),D组运动功能恢复最好。结论O-2A祖细胞移植可促进大鼠脊髓损伤后神经功能的恢复。 Objective To study the effects of O-2A progenitor cells transplantation on the repair of rat spinal cord hemisection.Methods O-2A progenitor cells from 1~2 days postnatal SD rats were dis-sociated,cultured and purified.Eighty SD rats were randomized to divide into four groups: the normal(group A,n=5);and the injury(group B,C and D,the recieved T9 laminectomy with spinal cord left-hemisection,each group n=25).The animals group B were recieved saline at injured spinal cord(the control group),those in group C were recieved 50% O-2A progenitor cell and 50% type 1 astrocyte(T1A) cell transplantation(O-2A+T1A) and in the group D were recieved purified O-2A progenitor cell transplantation(O-2A).Combined behavioral scores(CBS) and the Basso,Beattie,Bresnahan locomotor rating scale(BBB) of the rats were assessed by blinder observers 3,7,14,21,28 days after surgery.Results The CBS and the BBB scores in group A was 100 and 21 respectively.Certain neurological recovery in the hindlimb was observed in the injured groups 3,7,14,21 and 28 days after cell transplatation.CBS of the left hindlimbs was much higher than that in the right ones in the injured groups(P〈0.01),and the best recover was observed in group D.Conclusion Transplantation of O-2A progenitor cells might promote neurological recovery after rat spinal cord injury.
