抗Fas单克隆抗体诱导人肺癌细胞系A-549细胞凋亡 被引量:1

Induction of Apoptosis in Human Lung Cancer Cell Line A-549 by Anti-Fas Monoclonal Antibody

摘要 目的探讨抗Fas单克隆抗体诱导肺癌细胞凋亡的规律及在肺癌治疗中的意义。方法应用细胞形态学观察、琼脂糖凝胶电泳、流式细胞术检测抗Fas单克隆抗体对肺癌细胞A-549增殖周期的影响及对细胞杀伤作用的方式,并检测了A-549细胞表面Bcl-2的表达情况。结果抗Fas单克隆抗体有阻滞细胞周期、通过诱发凋亡而抑制肿瘤细胞生长的作用(P<0.05)。经抗Fas单克隆抗体处理后,A-549细胞表面Bcl-2蛋白表达差异无统计学意义(P>0.05)。结论抗Fas单克隆抗体可以诱导肺癌细胞系A-549细胞凋亡,抗Fas单克隆抗体诱导肺癌细胞凋亡与Bcl-2表达无关。 Objective To investigate the pattern of anti-Fas monoclonal antibody-induced apoptosis and its role in treatment of lung cancer.Methods Anti-Fas monoclonal antibody was tested for its effect on cell cycle and cell injury in lung cancer cell line A-549 with cell morphology,agarose gel elec-trophoresis and flow cytometry.Bcl-2 protein expression in A-549 cells was also examined.Results Anti-Fas mono-clonal antibody was able to block tumor cell proliferation cycle and trigger cell death by inducing apoptosis.The expression levels of Bcl-2 did not correlate with the apoptotic index after treat-ment with anti-Fas anti-body.Conclusion Anti-Fas monoclonal antibody can induce human lung cancer cell line A-549 apop-tosis.The biological effect does not correlate with endogenous Bcl-2 expression.
