摘要
目的研究白细胞介素-1β(IL-1β)及幽门螺杆菌(Hp)对人胃黏膜上皮细胞(GES-1细胞)增殖及超氧自由基阴离子()的生成和低剂量对GES-1增殖的影响。方法用3H-TdR掺入法测定cpm值及MTT比色法分析检测总活细胞数来估计GES-1细胞DNA合成及细胞生成,衡量GES-1细胞增殖情况。用细胞色素C还原法测定GES-1细胞、Hp产生水平。检测不同浓度Hp生长曲线及生成情况。结果IL-1β以剂量依赖性增加GES-1细胞DNA合成及细胞数量。IL-1β增加GES-1细胞产生。GES-1细胞能够自身产生。Hp生长情况与接种的菌量有关,Hp能够自身产生大量,其耐受的浓度明显高于胃黏膜细胞。结论IL-1β在调节GES-1细胞增殖中有重要作用。IL-1β涉及Hp感染胃黏膜细胞的高增殖反应和胃癌的致癌过程。IL-1β促生成的作用可能在胃相关疾病的防治中具有一定的意义。
Objective To study the effect of IL-1β and helicobacter pylori(Hp) on gastric epithe-lial cells proliferation and superoxide anion in culture.Metheds GES-1 cells were cultured with IL-1β.DNA synthesis of GES-1 cells was assessed by [3H]thymidine incorporation liquid scintillation counter and total viable cell numbers were analyzed by the MTT methed.The concentration of gen-erated by GES-1 cells and Hp were measured by ferricytochrome C reduction.Results IL-1β dose de-pendently increased DNA synthesis and cell numbers of GES-1 cells.Hp can generate spontaneous-ly,and endure higher concentration than GES-1 cells.GES-1 cells were also able to generate spontaneously.IL-1β promoted the proliferation of GES-1 cells by stimulating the generation of .Pro-liferation of GES-1 cells was induced obviously by the physical concentration of .Conclusion IL-1β plays an important role in the regulation of GES-1 cells proliferation.IL-1β involves in high-prolifera-tion and the carcinogenic processes of gastric mucosal cells infected by Hp.The effect of promoting generation by IL-1β may play a certain role in the prevention and remedy of stomach-related diseases
