摘要
利用BlastX将球毛壳菌(Chaetomium globosum)ESTs序列与GenBank的Nr数据库进行相似性搜索,获得含有丙酮酸脱羧酶基因(pdc)的2条ESTs(BP099214和BP113195),以此设计引物,克隆获得球毛壳菌pdc基因的cDNA及DNA序列。cDNA和DNA序列长度分别为1725和1829bp,编码574个氨基酸的多肽,蛋白质分子质量63.38ku。蛋白质家族预测其属于焦磷酸硫胺素TPP家族。BlastP相似性分析表明,PDC氨基酸序列保守性不高,与其相似性最高的是柄孢霉的PDC(Podospora anserina,CAD60727),相似性为79%。pdc基因的cDNA及DNA序列在GenBank上的登录号分别为EU304327,EU304326。
Two FASTs representing eDNA sequence of pyruvate decarboxylase gene (pdc) were obtained by blastX searching ESTs from Chaetomium globosum against Nr database of C, enBank. According to the sequence of ESTs representing pale ( BP099214 and BP113195), primers for PCR amplifying pdc gene were designed. The eDNA and DNA sequences ofpdc gene were cloned, with the length of 1 725 bp and 1 829 bp, respectively. The pdc gene encodes the protein of 574 amino acids with molecular mass of 63.38 kD. Protein family prediction indicates that this protein belongs to thiamine pyrophosphate (TPP) family. BlastP analysis reveals that amino acid sequences ofpdc gene from some fungal species share low similarity. The PDC from C. globosum shared the highest similarity with that from Podospora anserine, with a similarity of 79%. The eDNA and DNA sequences of pdc gene were accepted by GenBank ( accession numbers : EU304327, EU304326).
