嗜热链球菌活菌制剂保护剂及其工艺

摘要 为提高嗜热链球菌片剂的活菌存活率,延长其有效期,在扩大培养、制剂压片两个工艺流程分别加入保护剂,并利用数学统计方法进行了优化,得到最佳保护剂配方。首先利用Plackett-Burman设计法,对扩大培养时加入的8种材料及制剂压片时的12种材料的保护效果进行评价,筛选出影响嗜热链球菌存活率的两组重要因素。利用均匀设计法进一步得到重要因素的最适质量分数。验证试验表明:在扩大培养时加入0.5%蔗糖和3.5%可溶性淀粉对存活率有显著保护作用;在制剂压片时加入12.0%聚乙烯吡咯烷酮、25.0%可溶性淀粉、4.5%硫代硫酸钠、2.5%麦芽糖对存活率有显著保护作用。 An experiment was conducted to optimize protective agents of Streptococcus thermophilus in different technological processes in order to improve the survival rate of S. thermophilus, and prolong its period of validity. The important factors affected the survival rate were screened out by the Plackett-Burman design during cultivation and tablet forming of S. thermophilus. Further more, the regression equation of the important factors were obtained using uniform design. Results showed that the combination of 0.5% sucrose and 3.5% soluble starch was the optimal protective agents for expanding culture of S. thermophilus, while the combination of 12.0% polyvinylpyrrolidone, 25.0% soluble starch, 4.5% sodium hyposulfite and 2.5% maltose was the most effective protective agents for tablet forming of S. thermophilus.
