摘要
目的:观察六味地黄丸延缓5/6肾切除大鼠肾组织纤维化的作用。方法:SD大鼠随机分成正常对照组、模型组及六味地黄丸组;采用5/6肾切除术诱导大鼠肾小球硬化模型,于第5~8周分批处死大鼠。PAS染色观察大鼠残肾组织形态学改变;免疫组织化学方法分别检测肾组织中CollagenⅣ(Col-Ⅳ)/MMP-2的表达。结果:六味地黄丸组细胞增生明显轻于模型组,毛细血管开放较好,肾小管结构基本正常。Ⅳ型胶原在模型组中有较强的表达,尤其是在肾小管中染色很深,而在六味地黄丸组无论是肾小管还是肾小球着色均较浅,仅有少量表达;六味地黄丸组MMP-2表达强烈,染色明显深于模型组。结论:六味地黄丸可能通过促进MMP-2的表达,对抗Ⅳ胶原的过度沉积,减轻硬化程度,达到保护肾脏,延缓肾功能衰竭进程的积极作用。
Objective: To investigate the function of the Liuweidihuang Pills delaying renal fibrosis of the 5/6 of nephrectomy in rats. Method: SD rats were randomly divided into the normal control group, model group and Liuweidihuang Pills group, glomerulerosis model of rats was induced by 5/6 of nephrectomy and rats in batches at week 5,6,7,8 were sacrificed,the changes of the rest renal histomorphology of the rats was investigated by PAS staining: the expression of the Col-Ⅳ/MMP-2 was detected respectively in renal tissue with the immunohistochemistry method. Result:①In the Glomerular with PAS staining in model group the number of cells was increased significantly, the matrix increased and crescent generated. Mesangial area were widened and dyed deep, capillaries was opened badly or even blocked, renal capsule wall was thickened. While Liuweidihuang Pills group the cell proliferation was increased obviously lighter than that in the model group, capillaries opened better and the structure of renal tubule was basically normal.②In the model group, Ⅳ collagen had stronger expression, especially dyed deep in the renal tubular; while in the Liuweidihuang Pills group, whether renal tubuler or glomerulus were dyed light, Ⅳ collagen had a little expression. ③MMP-2 was expressed strongly in Liuweidihuang Pills group, and were obviously dyed deeper than that in the model group. Conclusion: Liuweidihuang Pills maybe through promoting the expression of MMP-2, antagonizing the excessive aggradation of type Ⅳ collagen and reducing the degree of glomerulosclerosis, plays active role of protecting the kidneys and delaying the process of renal failure.
