高效液相色谱法测定蝎龙接骨散中血竭素的含量

Determination of Dracorhodin in Xielongjiegu Powder by HPLC

目的:建立测定蝎龙接骨散活性成分血竭素含量的高效液相色谱法。方法:采用VP-ODS(250mm×4.6mm,5μm)色谱柱,以乙腈-0.05mol·L^(-1)磷酸二氢钠溶液(45:55)(用磷酸调pH为3.5)为流动相,流速为1.0ml·min^(-1),检测波长:440nm。结果:血竭素在9.59～47.94μg·ml^(-1)浓度范围内,峰面积与浓度线性关系良好,r=0.999 6,平均加样回收率为99.75%(RSD =0.99%)。结论:本法简便,准确,可用于蝎龙接骨散中血竭素的含量测定。

Objective： To establish an HPLC method for determination the content of dracorhodin in Xielongjiegu powder. Method： Samples were determined by HPLC on a VP-ODS column（250mm × 4.6mm,5 μm） with a mixture of acetonitrile -0.05mol· L^-1 Natrium bihydrogen phosphanate （ adjusted pH to 3.5 with phosphoric acid） （45：55 ） as mobile phase, the flow rate was 1.0 ml· min ^- 1 and UV detection wavelength was at 440nm. Result： The calibration curves of dracorhodin was linear from 9.59 to 47.94μg·ml^-1 （ r = 0.999 6）, the average recovery and RSD was 99.75% and 0.99%, respectively. Conclusion： The method is simple and accurate. It can be used for assay the content of dracorhodin in Xielongjiegu powder.
