摘要
用强度约为56μW/mm2的紫外线UV分别照射Page和Murcot原生质体30s、1min和5min,发现随着辐照时间的延长,原生质体活力趋于下降。Murcot的原生质体较Page的原生质体对UV敏感。辐射1min和5min的原生质体在培养过程中未能出现分裂,而是慢慢地发生质壁分离,最后破裂。而辐射30s的原生质体与对照都恢复分裂形成细胞团并长出愈伤组织。
UV,with the intensity of 56 μW/mm 2,was utilized to irradiate the protoplasts of Page tangelo(C.reticulata×C.paradisi cv. page )and Murcott tangor(C.reticulata×C.sinensis cv. murcott ).The result demonstrated that viabilities of the protoplasts decreased with the increased irradiation time.Murcott was more sensitive to UV than Page.The protoplasts irradiated for 1 minute and 5 minutes did not divide.Instead,most of them experienced plasmolysis and broke. Nevertheless, those irradiated for 30 seconds divided and developed into small calli.
