摘要
利用高粱EST序列和BAC克隆,以Primer 5.0程序设计了10对高丹草SSR引物,结果发现有3对引物在皖草2号高丹草的亲本间表现多态性,其中2对来自BAC的引物可用于遗传连锁图谱构建。除以上10对SSR引物外,再选取已公布的245对高粱SSR引物,以180个皖草2号(TX623A×S722)的F2代单株作为构图群体,构建了一个包括4个连锁群、33个SSR标记的高丹草遗传图谱,图谱覆盖基因组长度458.5cM,平均图距13.8cM。
Ten SSR primer pairs derived from EST and BAC in sorghum were developed using the software Primer 5.0 and 3 of them were polymorphic between Sorghum bicolor TX623A and S. sudanense S722, 2 of them were used for construction of genetic map. A genetic map of Sorghum bicolor × S. sudanense was constructed based on 10 SSR primer pairs developed and 245 publicly available SSR primer pairs for sorghum with a population of 180 F2 individuals derived from a cross between TX623A and S722. The map consisted of 4 linkage groups and 33 SSR makers. The map covered 458.5cM with average genetic distance of 13.8cM.
