TGF-β1在大鼠脊髓缺血再灌注损伤中的作用

摘要 目的探讨转化生长因子(TGF-β1)在脊髓缺血再灌注损伤中的作用。方法本研究采用脊髓缺血再灌注损伤模型,随机分为假手术组、脊髓缺血再灌注损伤组及TGF-β1抗体封闭组,每组32只,分别在术后6、12、24、48h后处死动物。用免疫组化检测TGF-β1、NGF、CD68蛋白表达。结果免疫组化结果显示缺血再灌注损伤后6hTGF-β1的表达开始增加(18.65±3.43),损伤后24h表达强度达高峰(39.18±0.34)。NGF蛋白表达与TGF-β1相似,6h开始增加(6.52±0.31),损伤后24h表达强度达高峰(20.08±4.34)。CD68蛋白的表达6h达到高峰(59.18±3.08),随后减低。TGF-β1抗体封闭后,TGF-β1蛋白的表达6h开始缓慢上调,NGF蛋白的表达12h开始增加以后表达平稳,CD68蛋白6h表达上调并持续高表达。结论上述结果提示脊髓缺血再灌注导致TGFβ1表达增多,可抑制炎性反应和促进NGF表达,从而有利于脊髓损伤修复。
