人趋化因子MIP-lα的克隆及其基因工程菌的构建

摘要 通过反转录聚合酶链式反应(RT-PCR)克隆并构建了去掉信号肽的人趋化因子巨噬细胞炎性蛋白(Mac-rophage inflammatory protein-1α,MIP-lα)基因的工程菌,并对该基因的结构进行了分析.结果表明,MIP-lα属于4Cys CC趋化因子,具有4个半胱氨酸,含有…CC……C……C……结构,该蛋白的4个保守半胱氨酸形成了两对功能必需的二硫键. The eDNA fragment encoding the human MIP-lα without signal peptide was obtained from mRNA of human T cells by reverse transcription-polymerase chain reaction (RT-PCR) and cloned into the pET28 vector. Computer-aided analysis showed that MIP-lα belongs to CC chemokines family and has two pairs of disulfide bond. The successful construction of MIP-lα gene expression vector lays the foundation for further research of MIP-lα protein function in resisting HIV infection.
