摘要
目的:观察兔骨髓基质细胞(MSC)的体外生长特性,为用于骨/软骨组织工程的功能细胞提供实验基础.方法:于胫骨近端抽取骨髓,分离MSC,原代培养,形成CFU-F,以1×10~4个细胞/cm^2传代培养5代,绘制细胞生长曲线,计算倍增时间,测定分裂指数和贴壁率.结果:第1~5代传代细胞生长曲线基本相同,第3代细胞倍增时间为24h,分裂指数最高达19%,12h内95%细胞贴壁.结论:在本实验培养条件下,MSC体外生长稳定,增殖速度较快,接种成活率较高,适应性强,可用于自体骨/软骨组织工程.
To investigate the growth characteristics of cultured rabbit marrow stromal cells (MSC). That will be the experimental basis of MSC used as the functional cells in bone/cartilage tissue engineering. Methods: Bone marrow was aspirated in the proximal part of tibia. MSC were separated for primary cuhur in which the CFU-F formed. Subculture was done for 5 passages consecutively, the initial cell density was 1×104cells/cm2. The following data were obtained; growth cure, cell double time, division rate and ad-henivve rate. Results: The growth curves of passage 1-5 were very similar. The double time of the third passage was 24 h. The highest division rate was 19‰. 95% of the subcul-tured cells were adhesive in 12 h. Conclusion: MSC show stable growth in vitro, relativey rapid proliferation and easily survive the subculture in present culture condition. MSC may be used in autogeneous bone/cartilage engineering.
